Tm calibration
For a melting curve assay, a melting temperature calibration file (Tm calibration file) may be required. This calibration file is a correction run to define the right offset for each Tm range. This offset is a deviation to correct for external factors influencing the temperature.
What are the ‘default instructions for use’?
In case no Tm calibration file is available, the ‘default instructions for use’ will be applied when performing an analysis. These ‘default instructions for use’ are the default Tm ranges as defined by the manufacturer’s instructions.
How can I a Tm calibration file?
In case of a melting curve assay, Tm calibration files are categorized under the lot section and can be added in 2 ways:
- Click the +-icon
- Choose USE A FILE or ADD MANUAL
- In case you choose USE A FILE
- Select a directory (a folder on your computer) and a Tm calibration file
- Click NEXT
- Assign the correct probes to the samples and select a CC
- Check the new Tm calibration values
- SAVE your results
- The Tm calibration file is now added to your assay
- In case you choose ADD MANUAL
- Choose a filename for the Tm calibration
- Click NEXT
- Change the Tm values for each target (when no values are added, the default Tm is used)
- Select the device you want to assign the Tm calibration to
- Click NEXT
- Check the overview of new Tm ranges
- SAVE the calibration
- The Tm calibration file is now added to your assay
How can I edit a Tm calibration file?
- Hover over the Tm calibration
- Click the EDIT ICON (pencil)
- You can now inactivate your Tm calibration file or set the file as default
- Click SAVE
- The Tm calibration is now inactive and appears grayed out
- If set to default, the Tm calibration will have a green checkmark
